Anaesthesia and gonad sampling in the European flat oyster (Ostrea edulis) ArchiMer

Controlled reproduction of the European flat oyster requires the development of tools adapted to this species, including a practical anaesthesia and gonad sampling protocol to facilitate sex determination and the verification of gametogenesis. Three replicate groups of 10 oysters (mean weight +/- SD: 29.9 +/- 8.5 g) were anaesthetised in 5 L containers using magnesium chloride designed either for laboratory (Flucka (R)) or agricultural (Dead Sea Work (R): DSW) use, at two concentrations (50 or 72 g L-1). Rearing turbot (Scophthalmus maximus) reaches the commercial scale. With the hatchery output increasing, more attention needs to be focussed on broodstocks performance both in terms of quantity and viability of eggs. The techniques of shifting the environmental conditions allow a production of eggs all year round. However, egg quality tends to be highly variable. This paper examines over a two-year period, the improvement in egg quality and its consequences on the production of larvae by a strict management of spawners.

Pacific oyster hatchery production remains highly variable, partially due to inconsistent gamete quality. However, little attention has been paid to oocyte quality in Pacific oyster compared to important vertebrate aquaculture species such as fishes. The aims of the present study were to identify criteria of oocyte and trochophore quality used as predictive tools for embryo developmental success at the D-larval stage. Twenty-six potential proxies of oocyte and trochophore quality were studied on 25 mature females, as determined using histology. Biological characteristics of European hake (Merluccius merluccius) sperm ArchiMer

Very little is known on European hake's reproductive biology and especially on biological characteristics of its sperm despite a growing interest in its aquaculture potential. This study reports, for the first time some hake sperm characteristics. After activation, the swimming phase lasts 3 min (8 min when activated with 50% sea water (SW) but lower initial velocity). The initial flagellar beat frequency (BF), velocity and percentage of motile cells decreased after 100 s. After 2 days at 4 degrees C, the mean Adenylate Energy Charge (AEC) level was 0.71. Sperm stored at 4 degrees C still showed motility after 9 days for 2 individuals. When cryopreserved, the motility recovery index of thawed spermatozoa ranged from 11.8% to 29.6%.

      The biology of gametes and larvae in brooding mollusc species remains little explored. The aim of this study is to present the results obtained in 2016, on biological characteristics of gametes and larvae of European flat oyster.       Measurements were carried out on 50 oysters, collected by divers in the Rade de Brest (Finistère, France), during the spawning period of this species. Gonad pH is acidic (6.58±0.54). The biology of cod reproduction is well described in the scientific literature. However, sperm biology and spermatozoa management are poorly studied in this species. Because of its recent farming expansion, a better knowledge of cod gametes is becoming especially useful. This work aimed at establishing tools to study sperm biology in cod, and also investigated the existence of changes in cod sperm quality during the spawning period. Ce rapport fait état d'une étude réalisée au laboratoire de Génétique et Pathologie de l'IFREMER dans le cadre de l'analyse d'épisodes de mortalité anormale survenue en écloserie privée de crevettes pénéides. Plusieurs hypothèses ont été avancées. La présence de Vibrio harveyi a amené à constituer une collection de 40 souches à partir desquelles une identification bactériologique a été effectuée. Elle montre en effet une dominance de souches appartenant à un groupe homogène de Vibrio harveyi. Pacific oyster Crassostrea gigas is one major species for aquaculture, and the development of breeding programs and the need for preservation of wild stock genetic resources prompted the need for larvae cryopreservation. The objective of the present study was to choose the most reliable protocol from several existing publications, to test its biological and practical limitations, and to adapt it to hatchery conditions. Cryopreservation of sea bass (Dicentrarchus labrax) spermatozoa in experimental and production simulating conditions ArchiMer

A sperm cryopreservation protocol adapted from turbot, was tested on sea bass using either 250-mu L straws or 1.5-mL cryovials. A dilution to 1/3 in Mounib's extender and a cooling rate of 65 degrees C.min(-1) allowed frozen sperm to recover an initial motility similar to that of fresh sperm at thawing; however, significant differences in motility (P < 0.001, n = 10 fish semen) were observed at further post-activation times, the motility decrease being faster in thawed sperm. At the experimental scale, triplicate inseminations of 2-mL aliquots (approximately 2 000 eggs) showed a significant fertility decay of thawed sperm compared to that of fresh sperm (P < 0.01, n = 12 fish semen) when a discriminating 35.10(3) spermatozoa to egg ratio was applied....

Cryopreservation of sperm in marine fish ArchiMer

Since the first work of Blaxter in 1953, fish sperm cryopreservation has been attempted on about 30 marine species. The present paper reviews the techniques used and the results published in these species. Particular attention is paid to the handling procedure of sperm before freezing, the problems of semen ageing and semen contamination with urine. The quality of frozen-thawed semen was evaluated using previously standardized biotests, such as a two-step motility activation technique adapted for the different species and fertilization assays using a discriminating insemination technique.
